How Arginine Influences Immune Surveillance

A study published in Cell shows that the amino acid arginine plays a direct role in the immune system’s ability to flag cancerous and virus-infected cells for destruction. Researchers at Rockefeller University found that when arginine was scarce, cells produced less of a protein called MHC-I, which acts as a “flag” for CD8 T cells — the immune system’s killer cells. Without enough MHC-I, tumors and viruses can more easily hide from the body’s defenses.

In mice with colon cancer, those given extra dietary arginine developed fewer and smaller tumors. In separate experiments, mice infected with influenza or a mouse-adapted SARS-CoV-2 virus fared better when arginine was supplemented: they lost less weight, survived in greater numbers, and showed reduced viral levels and lung damage. The supplemented dose was equivalent to about 10 grams per day in humans, a level the lead author says is relatively inexpensive and could one day complement existing treatments — if human trials confirm the effect.

The study uncovered a mechanistic link: low arginine causes ribosomes to stall during MHC-I production, so cells display fewer antigens on their surface. When arginine was restored, MHC-I levels recovered within hours. This suggests that the immune suppression seen in cancer and some viral infections isn’t just a byproduct of illness — it may be a direct consequence of arginine depletion driven by enzymes like arginase.

Why the Results Are Preliminary

Every expert contacted for this story stressed that the work remains confined to mice and cell lines. Dr. Sohail Tavazoie, the senior author, hopes to test arginine in high-risk flu patients, but no human trial has been conducted. Public health professor Jagdish Khubchandani noted that “unless there is a human trial, the findings have little value,” emphasizing that the study looked at highly controlled conditions that don’t reflect the complexity of real-world disease or diverse patient populations.

Safety Questions Loom Large

The 10-gram human-equivalent dose is not trivial. The NIH reports that 9–30 grams per day can cause gastrointestinal discomfort and a slight drop in blood pressure. Surgical oncologist Anton Bilchik warned that while most healthy people might tolerate the dose, it could be risky for those with kidney or liver disease. Pregnant individuals and those with cardiovascular conditions should also avoid such high intake without medical oversight.
